My experience with this is that you need to have at least half as many interfaces as you have EDS cards. If you have 8 cards, you'd need interfaces on cards 1, 3, 5, and 7, with a word clock generator.
Having only one interface may work, but there are synchronization errors that lead to time-outs in the PSCL. This causes the PARIS app to become unresponsive for a second or so sometimes, and submixes will just not play sometimes.
With 4 cards, I'd have an MEC on card one, and a 442 on card 3 with a word clock generator. The Hosa one works great, and it's cheap.

You'll need 2 EDS cards to run 2 C-16s and 32 tracks without a submix. This is my set up with 1 MEC. I slave (hook up the EDS cables) per the manual.
Each EDS card can run 1 C16 and a 16 channel submix.
I only have 1 MEC with 2-8ins so I only record 16 at a time, but I mix via 2 EDS cards and 2 C16s up to 32 tracks.
